We are modeling motor neuron disease (MND) using Drosophila and mice expressing mutant TDP-43, a DNA/RNA binding protein central to the pathogenesis of MND. This work was initiated in collaboration with Dr Marc Freeman and Dr Robert H Brown at UMass Medical School. Using our fly model, which mimics more closely the human condition than previous models, we identified novel suppressors of TDP-43 toxicity. We are working to study these suppressors in mammalian systems using in vitro cultured primary motor neurons. We are also performing a range of in vivo studies in mice and have generated a novel TDP-43 mouse that is hoped will more closely recapitulate the human condition at the molecular level.
